Tara is Sookie's best friend and the bartender at Merlotte's. As a result of being raised in an abusive home, Tara has difficulty trusting others. Her propensity for driving away those that are close to her leads her to be scammed by Miss Jeanette; a woman who offers to exorcise a demon in Tara that she claims is the source of the problem. Towards the end of the first season, Tara is arrested for driving under the influence and is taken under the wing of Maryann Forrester (despite Maryann and Daphne being the cause of Tara's collision with a tree). While under her care, Maryann begins manipulating Tara so that she can control the people of Bon Temps. While at Maryann's, Tara meets "Eggs" Benedict Talley, who she gradually begins a relationship with during the second season. Before Sookie leaves for Dallas, she's invited to move into Sookie's house; Eggs and Maryann also move into the house shortly there after.

His flamboyant personality leads to numerous confrontations with a trio of homophobic customers at Merlotte's in the first season, and he's shown spending his time away from the bar partying and creating webcam pornography in his house. He sells vampire blood (a human narcotic known by the street name "V," within the series) to Jason, but refuses to sell him any more after Jason takes too much at once. After Eddie (his source for V) goes missing, he is kidnapped by Eric Northman in the final episode of the first season. In season two, he's revealed to be locked in a dungeon with Royce Williams, a customer he had previously kicked out of Merlotte's. When Sookie learns of Lafayette's imprisonment, she bargains with Eric for his freedom and takes Lafayette back to his home.

Jason Stackhouse is Sookie's dim-witted brother and a road crew supervisor known for bedding the women of Bon Temps in his spare time. During the first season, he is the prime suspect in the murders of three women he is known to have been involved with before their deaths: Maudette Pickens, Dawn Green, and Amy Burley. After losing Amy, he turns himself in for their murders and (while in jail) is recruited by an anti-vampire church: the Fellowship of the Sun. During the second season, he feels he has found a purpose with the church; asking for a "sign" of what God wants him to do. He believes that sign comes when Sookie refuses their uncle Bartlett's inheritance money, giving it all to Jason. He then attends a leadership conference with the Newlins, the leaders of the Fellowship of the Sun. Although he and Sookie spend sparse time together, Jason will do almost anything in his power to defend and protect her.

Detective Andy Bellefleur is a detective in Bon Temps who is regularly disrespected by the town's citizens. In the first season, he is assigned the investigation of the murders that occur during the first season, and is set on arresting Jason Stackhouse for the crime; even when the evidence doesn't support this. Though he is initially praised for his actions, he begins drinking heavily once the true killer's identity is revealed. During one of his binges at Merlotte's, the body of Miss Jeanette is left in his car; making him a suspect in her death. Because of his nearly constant state of impairment during the second season, Sheriff Dearborne takes his badge, his gun, and suspends him from official police work.

For the majority of the first season he is bossed around by his mother, but stands up to her after Jason leaves his comfort zone to be with Amy. Sheriff Bud Dearborne is the town sheriff and an old friend of Sookie's parents. While he is well respected in the town of Bon Temps, he goes out of his way to show his distaste for vampires and vocally objects to Sookie's relationship with Bill. He doesn't believe Jason is the murderer in the first season, and regularly scolds Andy for not being fair and unbiased to suspects (despite the blatant prejudice he shows to Bill himself). Lettie Mae Thornton is Tara's abusive, alcoholic mother. After attacking and driving Tara away in a drunken stupor during the first season, she unexpectedly shows up at Adele Stackhouse's funeral and acts unusually composed. She convinces Tara to fund an exorcism to expel a demon that she is convinced is responsible for the terrible things she has done, and chooses to corrupt her because of her closeness with Jesus. While the fake exorcism appeares to have the desired effect, it has become clear that she can be just as cruel sober. After Miss Jeanette gets her heart ripped out, Lettie Mae is called to the police station for questioning in season two. While there, Tara tells her that the exorcism is fake but she doesn't believe it. Maryann points out the harmful things she's done to Tara when she and Lettie Mae first meet. On Tara's birthday, Lettie Mae asks Sam to deliver a present to her that Maryann later discards.

Bill Compton is a vampire with whom Sookie falls in love, and is a main protagonist in the series. Turned during the civil war against his will, Bill shows a remarkable and unusual compassion for human life. He is also outwardly expressive of his emotions and holds on to memories of his past human life; behaviors that are unique amongst his vampire peers. Many times, Bill risks his life and standing in vampire society to protect Sookie from the dangers their relationship has brought on. During the second season Eric's interest becomes a romantic attraction. This is demonstrated when he tricks her, under the false pretense that he could not heal, into sucking silver bullets out of his body in order for her to drink his blood. It is then revealed that having his blood in her would cause Sookie to become attracted to Eric in addition to allowing him to monitor her emotions and location.

Maryann Forrester is a wealthy, mysterious "social worker" and maenad. Before the events of the series, she has a brief encounter with Sam Merlotte when he is 17; the two become romantically involved and Sam steals some of her belongings. After arriving in Bon Temps, Maryann's standing in the middle of the road forces Tara to crash her car into a nearby tree. Tara is arrested for driving while under the influence, but Maryann posts her bail (when Lettie Mae will not), invites her into her "home", and introduces her to "Eggs" Benedict. Maryann has an assortment of strange abilities that includes: the ability to influence or control the behavior of others, force shapeshifters into changing shape, is immortal, and can transform her hands into beast-like claws which exude a powerful neurotoxin. She uses this last ability to attack Sookie; who Bill takes to Eric in order to keep her from dying. During the second season (with Bill and Sookie out of Bon Temps), she uses her mind control to take over the town, holds orgies, and attempts to sacrifice Sam. She initially uses Daphne to bait Sam, but eventually has Eggs kill Daphne and dump her body in the meat freezer at Merlotte's (implicating Sam as her killer). She then unleashes the whole town to find Sam, who just escaped jail.

Eddie Gautier is the vampire who supplies Lafayette with V in the first season. Before the series takes place, his relationship with his wife falls apart because he's gay. After he is kicked out of their house, his physical appearance makes him unable to form a relationship with any men. He becomes a vampire to improve his dating life, but he continues to lead a sedentary life as he is unable to glamor others. Instead, he spends most of his time watching television and supplying Lafayette with his blood in return for sexual favors (under the misconception that Lafayette has genuine affection for him). He is kidnapped by Jason and Amy to use as a blood supply, and is later staked by Amy after Jason attempts to free him. During the second season, Jason has a nightmare where he wakes up to find Eddie laying with him in bed.

Maudette Pickens is a local woman who videotapes her sexual encounters with various men, including Jason Stackhouse and a vampire named Liam. While having sex with Jason in the first season, he grabs her by the neck (imitating the way she's treated in a video with Liam), and she deceives him into thinking that he's choked her to death. Later that night, after Jason leaves her home, Drew Marshall kills her and steals her collection of tapes, with the exception of Jason's, which the Bon Temps police discover.

Mack and Denise Rattray are a couple who attempt to drain Bill Compton of his blood in the early episodes of season one. After Sookie attacks them, they threaten to get her back and nearly beat her to death in the parking lot the next night. Bill kills them before they can succeed; attempting to make their deaths appear as though they are the result of a tornado descending upon their house.

Bartlett Hale Adele's brother and Sookie's great-uncle. In the first season, it's revealed that he molests Sookie as a child, and later becomes estranged from the rest of the family after Sookie tells her grandmother what happened. After he appears at Adele's funeral, Sookie confides all this to Bill. Bill later tracks him down and kills him in an act of revenge. In his will, he bequeaths Sookie $11,000, all of which she gives to Jason.

Malcolm, Liam, and Diane are vampire acquaintances of Bill Compton who form a nest with one another and come to live in Bon Temps during the first season. None of them have any interest in mainstreaming and have little regard for life. Malcolm is the oldest of the three, is indeed older than Bill, and assumes a leadership role in the nest. He likes to party and has a human lover named Jerry. Sookie discovers, telepathically, that Jerry has Hepatitis D and is trying to infect the nest. Liam pays Maudette Pickens a large sum of money to have sex with her, and was videotaped in the act. Before the events of the series, Diane sleeps with Bill shortly after she's turned and has been trying to seduce him since. All three vampires are killed in a fire set by Royce Alan Williams and his associates.

Nancy LeGuare is first introduced in season one as Miss Jeanette, she works as a drugstore clerk but also performs fake "exorcisms" on the side to support her family. Her body was discovered at the end of the last episode of season one in Andy Bellfleur's car. In the first episode of season two it is revealed her heart has been cut out and in a later episode it can be seen that Maryann served her heart to her party guests. It's revealed that she had scratches on her back similar to Sookie's; Maryann being the creature that attacks both women.

Royce Alan Williams is a Bon Temps local who burns down the house that Diane, Malcolm, and Liam are living in; also killing their guest, Neil Jones, in the process. Typically depicted with two friends, they not only have an open distaste for vampires but send back food prepared by Lafayette at Merlotte's (implying that they'll catch AIDS if they eat it because he's gay). He is captured by Eric Northman for killing the three vampires. He confesses to Lafayette that he's been a screw-up most of his life, including a sexual adventure with one of his cousin's girlfriends and a homosexual encounter in camp. During an escape attempt, he is killed by Eric.

She is revealed to have strange claw marks on her back that are similar to the ones found on Miss Jeanette and inflicted on Sookie. Eventually, Daphne is found to have been working for Maryann, and betrays Sam to her mistress. She is later murdered by Eggs, at Maryann's behest.

Luke McDonald was a member of the Fellowship of the Sun. He was both a friend and an enemy to Jason Stackhouse, and ultimately led a suicide mission against the Dallas Vampires.
Luke grew up in Odessa, Texas. During high school, he played football and brought his team to the state finals. After graduating, he spent most of his time with his girlfriend, Katie, who was attending Sam Houston State University, and partying with his high school friends. Instead of enrolling in college, he enlisted in the army, and was denied entrance when the entrance physical revealed that he had a bad knee.
Three years later, he signed up to join a training camp for the Fellowship of the Sun, an anti-vampire church. On the bus ride there, he bonded with Jason Stackhouse, though he later became jealous of the amount of attention he was receiving. After the camp ended, Luke was selected to join the Soldiers of the Sun. While he was initially thrilled, he was worn out by the strenuous activity that the training required. During his training, he posted information on his blog regarding vampires in the media and on his girlfriend, Katie, who is implied to be against vampire rights as well. Later, he reveals that he feels far removed from his life back home, as he missed Katie’s graduation, and the two of them eventually break up when she moves to California to take a job in Pharmaceuticals. At camp, Luke tells Jason that the only reason that Sarah likes him is because she wants to sleep with him, making his resentment clear to everyone.
Luke and Jason are chosen to work on a special project: a platform for a vampire to meet the sun. He repeatedly states that he feels joyful to be part of the upcoming war on vampires. However, his enthusiasm is replaced by shock when Jason joins the vampires to help save his sister from getting killed by the Newlins. While it initially appears that Luke may be leaving the Fellowship, he shows up at the Dallas vampire headquarters later that night with a bomb strapped to his chest. He tells Jason to leave, and blows himself and the building up.

Godric is a 2,000-year-old vampire who looks no older than a teenager. It is revealed that he is the oldest vampire in the New World. He has tattoos from Roman times. When he goes missing, Eric sends Sookie and Bill to the Dallas area to look for him. Godric takes the blame for the "PR disaster" stemming from his decision to let himself be captured by the Light of Day Institute to be a sacrifice. He relinquishes his title as sheriff of Area 9 and commits suicide by meeting the sunrise on the roof of the Dallas hotel, while a tearful Sookie looks on.
